    Background: 
    Aurora A (AIK) is a cell cycle-regulated Ser/Thr protein kinase that is overexpressed in many tumor cell lines. Phosphorylation of Aurora A at Thr288 within the kinase activation loop results in a significant increase in its activity and may target the protein for proteasomal degradation during mitosis . The closely-related kinase Aurora B (AIM1) has been implicated in multiple mitotic events, and siRNA silencing of Aurora B expression results in reduced histone H3 phosphorylation, aberrant chromosome alignment/segregation, and altered survivin localization.

    Western Blot: Aurora B Antibody - Left: Analysis of HeLa cells treated with 100ng/ml nocodazole for 18 hours was probed with this antibody. Nocodazole is a microtubule polymerization inhibitor which induces cells to halt at the G2/M phase and also induces Aurora B expression. this antibody binds strongly to Aurora B at 38 kDa. Right: Blots of recombinant human Aurora A, B and C were probed with this antibody.

    Immunocytochemistry/Immunofluorescence: Aurora B Antibody - HeLa cell cultures were stained with this antibody(green). Localization of Aurora B is cell cycle phase dependent. First, Aurora B staining was seen in chromosome arm in cells in prophase (green small dots). During anaphase, Aurora B localized in midzone and then concentrated in midbodies between the two daughter cells during telophase. It is therefore a useful marker of midbodies and dividing cells. Cells were counterstained with chicken polyclonal antibody to Vimentin (red). Blue is a DNA stain.

    Immunocytochemistry/Immunofluorescence: Aurora B Antibody - Analysis of HeLa cells stained with antibody to aurora B kinase, dilution 1:1000 in red, and costained with chicken pAb to vimentin, dilution 1:1000 in green. Blue is DAPI staining of nuclear DNA. this antibody produces strong staining associated with chromosomes in prophase (A), the centromere in prometaphase and metaphase (B), the central mitotic spindle in anaphase (C), and midbodies between the two daughter cells during telophase and beyond (D). The vimentin antibody stains the intermediate filament network in these cells.
