N6-Methyladenosine (m6A) is an abundant modification found in mRNA, tRNA, snRNA, as well as long non-coding RNA, in all species. RNA adenosine methylation is catalyzed by a multicomponent complex composed of METTL3/MT-A70, METTL14, and WTAP in mammals. METTL3 & METTL14 are responsible for the methyltransferase activity of the complex, and WTAP mediates substrate recruitment.

Dot Blot - Anti-N6-methyladenosine (m6A) antibodyTo demonstrate the specificity of AT15003 against N6-methyladenosine (m6A), a Dot Blot analysis was performed using synthetic oligonucleotides containing different modified bases. 150 and 50 pmol of the oligonucleotides were spotted on the membrane. The antibody was diluted 1/400 in TBS-T containing 10 % skimmed milk and 1% BSA. AT15003 shows a high specificity for the oligonucleotide with the N6-methyladenosine modification.
