    This DAPI Staining Solution is a carefully optimized staining solution suitable for almost all common cell and tissue nucleus staining. Can be directly used for nuclear staining of fixed cells or tissues.

    DAPI, The molecular weight is 350, CAS Number 28718-90-3.

    DAPI is a blue fluorescent dye that can penetrate cell membranes. After binding with double stranded DNA, it can produce fluorescence that is more than 20 times stronger than DAPI itself. Compared to EB, the staining sensitivity for double stranded DNA is many times higher.

    DAPI is commonly used for staining double stranded DNA in the nucleus, and after staining, it is observed under a fluorescence microscope or detected by flow cytometry.

    The maximum excitation wavelength of DAPI is 340nm, and the maximum emission wavelength is 488nm. After DAPI binds to double stranded DNA, the maximum excitation wavelength is 364nm and the maximum emission wavelength is 454nm.

    Protocols
    1. After fixing the cell or tissue sample, wash it appropriately to remove the fixative. If immunofluorescence staining is required later, immunofluorescence staining should be performed first, and then DAPI staining should be performed according to the subsequent steps. If no other staining is required, perform DAPI staining directly.

    2. For adherent cells or tissue sections, add a small amount of DAPI staining solution and cover the sample. For suspended cells, add at least 3 times the volume of staining solution to the sample to be stained and mix well.

    3. Leave at room temperature for 3-5 minutes.

    4. Discard DAPI staining solution and wash 2-3 times with TBST, PBS, or physiological saline, each time for 3-5 minutes.

    5. Observe directly under a fluorescence microscope or under a fluorescence microscope after encapsulation. When cells undergo apoptosis, the nuclei of apoptotic cells can be seen to be densely stained or fragmented with dense staining.
