    The ECL Western Blotting Substrate is a highly sensitive, nonradioactive, enhanced luminol-based chemiluminescent substrate for easy detection of horseradish peroxidase (HRP) on immunoblots. This ECL Western Blotting Substrate offers superior signal and clear background. Blots can be repeatedly exposed to X-ray film to obtain optimal results or stripped of the immunodetection reagents and re-probed.

    DETECTION PROCEDURE:

    1. Mix Detection Reagents A & B at a 1:1 ratio.
    2. Immediately add the mixture to the blot. Use 0.125 ml working solution per cm2 of membrane.
    3. Incubate the blot for about 1 minute at room temperature.
    4. Drain excess reagents.
    5. Cover blot with a clear plastic sheet protector or clear plastic wrap.
    6. Expose blot to X-ray film.

    ECL chemiluminescence solution is used to detect antibodies that directly or indirectly label horseradish peroxidase (HRP) and its associated antigens. The detection principle is shown in Figure 1. After electrophoresis, the protein is transferred to the imprinted membrane, and the target protein on the membrane is bound by the primary antibody and the HRP labeled secondary antibody. After washing the film, incubate the film with ECL chemiluminescence solution at room temperature for several minutes, and the target protein can obtain clear bands on the X-ray film.
